I do follow the C-32Bs and COMCO's but have no idea on the serials as i actually don't look at them.

Aha, an interesting link after a bit of digging this morning (strangely found on a flight simulation forum, not sure where it was taken from!)
02-5001 and 00-9001 are the same airframe, as is 99-6143. This may, or may not be one of the two Comco birds!

COMCO B757's do not have an AR Receptacle but the C-32Bs do have. So easy to tell between both if you have a front on/slightly elevated view.
Plus COMCO's are merely known as B752's and not C32. They are completely different aircraft and mission albeit both types are self sufficient when operating in dodgy locations.

The plain white C-32B's 24452 and 09001 are often seen overseas, while 25001 and 96143 assigned to the 486th FLTS at Eglin FL stay in the US. 25001 and 96143 both carry US Air Force titles, perhaps they have now joined the other two C-32B with the 150th SOS at McGuire NJ as 96143 has started to be seen around Europe.
The COMCO Boeing 752's N226G and N610G make tech/fuel stops at Manchester. I think they have also been seen at Brize Norton, Bournemouth and Shannon.
Three other Boeing 752's ... N144DC, N440AN and N701TW are also registered to the US Air Force or L-3 Capital LLC.

The COMCO's rarely make ground for more than a quick turn around and Shannon is one of the primary spots for this.
Although we know what the C-32B's are normally used for, i don't know of anyone yet (not in on the programme) that knows exactly what or who the COMCO's transport around. They end up in places you would expect the usual agency to be playing in but i think they are used for more than one agency, most probably NSA or the like.
Would be nice to see what they are like internally... if they have a different layout than the normal B757s and if they carry around a large pax number or small. I know that on one occasion whilst at Brize, some of those onboard had US Military cammo on.....

No idea how much they use Shannon i was just stating that Shannon "is one of the primary spots", not "the" most used location.
The SOS/SOF now comes under the 108th as far as i am aware.
The Comcos seem to have a mix of military and civilian onboard.

In case anyone's interested, am sitting sipping a cool beer with toes in the pool near Eglin and mode-S code AE0433 - which may be 25001 - just arrived. Unfortunately it's dark, so couldn't read any serial.
Google Earth of Eglin shows a C-32 on its own parking spot ready for action but the imagery is 2007, so maybe if they're no longer based it'll be at the terminal next to an American Eagle Embraer 145.
Taliking of COMCOs, N610G departed Mobile Downtown (they seem to hang out there) yesterday afternoon; according to flight trackers, it went to Denver and today to Spokane.

This is another one of those mysterious Boeing 757s apparently used by the US State Dept Foreign Emergency Support Team (FEST); apparently it carries the 'tail number' 86006 which may or may not be genuine. Has been listed as c/n 27204, which would make it ex B-27007 of Far Eastern Air Transport, but c/n 27204 has been flying with Angkor Airways as XU-AKB There are at least four 757s that could be this plane--c/n 25493/523 ex N84WA, 25494/611 ex N987AN, c/n 25491/511 (N226G) and c/n 29304/870 (N610G).
It is actually c/n 25494/611, which has carried serials 98-6006, 99-6143, 00-9001, and 02-5001 interchangeably.

Interesting that this has just popped back up to the top as I saw what I believe was "25001" arriving at NAS North Island on Sunday, 2nd Oct. Was calling "Alton? 64" and was a night-stopper. In an effort to settle the debate over the current serial of the Eglin C-32B a photo was required. Unfortunately, on the evening in question, it departed after sunset, so in the grainy photos department, this is super-grainy and fails to ID the airframe.

I assume those engines are RB211s? They look more elongated than I'd expected.
Whilst the camera operator took the photos I was looking through bins and it had U S AIR FORCE titling and the national insignia on the rear fuselage.
During the day it's sat in the usual place as per Google Earth at Eglin.
